Definition and Calculation
The lethal dose is typically expressed in milligrams per kilogram of body weight (mg/kg). A low LD50 value means that a substance is lethal even in small quantities and is therefore considered highly toxic. A high LD50 value, on the other hand, indicates lower toxicity.
- LD1: The dose at which 1% of a population dies
- LD50: The dose at which 50% of a population dies (most common reference value)
- LD100: The dose at which 100% of a population dies
These values are typically determined through animal studies and subsequently extrapolated to humans using safety factors. Today, alternative methods that do not involve animals – such as in vitro tests and computer-based models – are increasingly being used.

Therapeutic Window and Safety Margin
In pharmacology, the therapeutic window (also known as the therapeutic index) is a key concept. It describes the range between the dose that produces the desired therapeutic effect and the dose that causes toxic or lethal effects. A narrow therapeutic index – as seen with certain heart medications or anticoagulants – requires particularly careful dosing and regular medical monitoring.
