What Are Endocannabinoids?
Endocannabinoids are lipid-based signaling molecules produced naturally by the human body. They bind to cannabinoid receptors and regulate a wide range of physiological processes. The term combines endo (from Greek, meaning within) and cannabinoid, indicating internally produced compounds with cannabinoid-like activity. The two most well-known endocannabinoids are anandamide (AEA) and 2-arachidonoylglycerol (2-AG).
The Endocannabinoid System
The endocannabinoid system (ECS) is a complex signaling network in the human body consisting of three main components:
- Endocannabinoids – the endogenous ligands (signaling molecules)
- Cannabinoid receptors – primarily CB1 and CB2 receptors
- Enzymes – responsible for synthesizing and degrading endocannabinoids (e.g., FAAH, MAGL)
CB1 receptors are predominantly found in the central nervous system, whereas CB2 receptors are mainly expressed in immune tissues and peripheral organs.
Biological Functions
Endocannabinoids act as retrograde messengers: they are produced on demand and travel backward across synapses to modulate the release of other neurotransmitters. They are involved in many essential body functions, including:
- Pain perception – modulation of pain signals in the nervous system
- Mood and emotional well-being – regulation of anxiety, stress, and pleasure
- Sleep – supporting healthy sleep onset and maintenance
- Appetite and metabolism – control of hunger and energy balance
- Immune function – modulation of inflammatory responses
- Memory and learning – involvement in synaptic plasticity
- Motor coordination – influence on movement and motor control
Anandamide and 2-AG in Detail
Anandamide (AEA)
Anandamide takes its name from the Sanskrit word ananda, meaning bliss, and is often referred to as the bliss molecule. It preferentially binds to CB1 receptors and influences mood, pain sensitivity, and the experience of anxiety. Anandamide is rapidly broken down by the enzyme FAAH (fatty acid amide hydrolase).
2-Arachidonoylglycerol (2-AG)
2-AG is the most abundant endocannabinoid in the brain and binds to both CB1 and CB2 receptors. It plays a key role in regulating inflammation, immune responses, and neuronal excitability. Its degradation is primarily carried out by the enzyme MAGL (monoacylglycerol lipase).
Medical Relevance and Therapeutic Context
Dysregulation of the endocannabinoid system has been linked to various conditions, including chronic pain syndromes, anxiety disorders, irritable bowel syndrome, fibromyalgia, and migraine. The concept of clinical endocannabinoid deficiency (CECD) describes states in which endogenous endocannabinoid production is insufficient to maintain homeostasis.
Plant-derived cannabinoids such as cannabidiol (CBD) and tetrahydrocannabinol (THC) from the cannabis plant also interact with this system and are increasingly being used therapeutically in modern medicine for indications such as neuropathic pain, chemotherapy-induced nausea, and certain forms of epilepsy.
Lifestyle Factors Affecting the Endocannabinoid System
Several lifestyle factors can influence the activity of the endocannabinoid system:
- Exercise and physical activity – aerobic exercise has been shown to elevate anandamide levels (the so-called runner's high effect)
- Diet – omega-3 fatty acids serve as precursor molecules for endocannabinoids
- Stress – chronic stress can burden the ECS and promote dysregulation
- Sleep – adequate sleep supports healthy ECS functioning
