The azo group is a chemical functional group defined by the structure -N=N-, in which two nitrogen atoms are connected by a double bond. It is the defining feature of azo compounds and azo dyes, one of the largest and most commercially important classes of synthetic dyes. These compounds are found in many everyday products, from food and cosmetics to textiles and pharmaceuticals.
Occurrence and Uses
Azo dyes are widely used across various industries:
- Food: As synthetic colorants in beverages, confectionery, snacks, and other processed foods. Well-known examples include Tartrazine (E102), Carmoisine (E122), and Allura Red (E129).
- Cosmetics: In lipsticks, hair dyes, and other beauty products as coloring agents.
- Textiles: Widely used to dye fabrics and other materials.
- Pharmaceuticals: Occasionally used as excipients in tablets or capsules for identification purposes.
Health Considerations
Intolerances and Allergic Reactions
Certain azo dyes in food can trigger adverse reactions in sensitive individuals, including:
- Skin reactions such as urticaria (hives) or contact dermatitis
- Respiratory symptoms resembling asthma
- Gastrointestinal complaints
A well-documented example is tartrazine intolerance. People with aspirin sensitivity or asthma may also show cross-reactivity to certain azo dyes.
Hyperactivity in Children
European regulatory authorities, including the EFSA and the UK Food Standards Agency (FSA), have reviewed evidence suggesting that a combination of certain azo dyes and the preservative sodium benzoate may affect the behaviour and attention of children. As a result, food products in the EU that contain specific azo dyes must carry the warning label: May have an adverse effect on activity and attention in children.
Metabolism of Azo Dyes in the Body
In the human body, azo dyes can be broken down by intestinal bacteria or liver enzymes through a process called azo reduction. This process releases aromatic amines, some of which are considered potentially harmful or carcinogenic. The EFSA therefore regularly reassesses the safety of approved azo food colorants.
Regulation in the EU
In the European Union, only a limited number of azo dyes are approved for use in food. Under EU Regulation (EC) No 1333/2008 on food additives, all permitted colorants must be declared on the ingredient list by their name or corresponding E-number. Azo compounds capable of releasing carcinogenic aromatic amines are prohibited from use in both food and textiles.
