What is Pantoprazole?
Pantoprazole is a widely used medication belonging to the class of drugs known as proton pump inhibitors (PPIs). It is available both by prescription and, at lower doses, over the counter. Pantoprazole is one of the most commonly prescribed medications worldwide and is used to treat a range of conditions affecting the upper gastrointestinal tract by reducing the amount of acid produced in the stomach.

Pantoprazole is indicated for the following conditions:
- Gastroesophageal reflux disease (GERD): Reduces acid reflux and relieves symptoms such as heartburn and regurgitation.
- Gastric ulcer and duodenal ulcer: Promotes healing of the stomach lining and prevents recurrence.
- Zollinger-Ellison syndrome: A rare condition characterized by excessive acid secretion due to a gastrin-producing tumor.

Mechanism of Action
Pantoprazole acts as an irreversible proton pump inhibitor. After absorption, the drug is activated in the acidic environment of the stomach wall and binds permanently to the H+/K+-ATPase enzyme — the proton pump located in the parietal cells of the gastric mucosa. This enzyme is responsible for secreting hydrochloric acid into the stomach. By irreversibly blocking this pump, pantoprazole produces a long-lasting reduction in gastric acid secretion. The effect persists until new pump molecules are synthesized by the gastric mucosa.
Dosage and Administration
Pantoprazole is available in several formulations, most commonly:
The standard adult dose ranges from 20 mg to 40 mg once daily, depending on the indication and medical advice. Tablets should be taken whole, without chewing, approximately 30 to 60 minutes before a meal. This ensures that the enteric coating remains intact and the active ingredient is released in the small intestine rather than the stomach.

- Antifungal agents (e.g., ketoconazole): Absorption may be impaired due to altered gastric pH.
